Abstract

How do we perceive the scientific cause of arthritic disease
Arthritic disease is one of the immune system concerning malfunctions to the arachidonic cascade resulting in excesses in production of members of the prostanoid family of hydrocarbons. Pharmacological action of Joint Massage Oil (NBS)
I've witnessed NBS reducing inflammation so many times, the effect has got to be fact. Logical explanation is more difficult as I believe NBS acts in a novel way. These are my theorisations based on scientific study and 14yrs of experience with arthritis sufferers.
"NBS may block the production of key hormones that are used to carry local messages in the inflammation process involving prostaglandins, cyclooxygenase and arachidonic acid".
I believe a natural chemical in NBS blocks the binding of arachidonic acid in the local cyclooxygenase active sites so the pain reseeds and inflammation is reduced.
Here's how
We know prostaglandin synthase is a complex dimer* of identical subunits that acts at the site of pain and inflammation. Each subunit has a small carbon-rich knob. These knobs anchor the complex to the membrane of the endoplasmic reticulum. The cyclooxygenase active site is buried deep within the protein, and is reachable by a tunnel that opens out in the middle of the knob. This acts like a funnel, guiding arachidonic acid out of the membrane and into the enzyme for processing.
It is reasonably possible that ionic bonding by constituents of NBS at the entrance of the cyclooxygenase active site may inactivate the enzyme.
*A dimer has bifold symmetry allowing the recognition helix of the second protein sub-unit to make the same groove binding interactions as the first.
Background
Prostaglandins are the molecular messengers used to signal pain and inflammation. There are different prostaglandins, but all are created from a common precursor molecule. They are delivered throughout the body by the blood in response to injury, infection or metabolic disorder. They act only in the surrounding area before they are broken down. Occasionally the event also damages the DNA controlling expression in the enzyme triggering autoimmune disease.
Cyclooxygenase performs the first step in the creation of prostaglandins. It adds two oxygen molecules to arachidonic acid, beginning a set of reactions that will ultimately create a host of unusual molecules. Arachidonic acid is an unsaturated fatty acid with several double bonds that form rigid kinks in the carbon chain. There are different cyclooxygenases for different purposes. COX-1 & COX-3 are built in many different cells to create the prostaglandins used for basic housekeeping messages throughout the body. COX-2 enzyme is built only in special cells and is used for signaling pain and inflammation. If COX-1 is targeted as well in the case of conventional treatment, this can lead to unpleasant complications, such as stomach bleeding and other side effects.

Prognosis
Prostaglandin synthase has C20,H32 hydrocarbon building blocks. Several components of NBS have C5,H8 phenolic antioxidants.4(C5,H20)=C20,H32 + antioxidants. A hydrocarbon structure bridges the Cyclooxygenase funnel and the antioxidants bind the oxygen release of cox-2 signalling the mechanism for normal break down of the flawed cell and thus promoting natural healing.
Arthritic Degeneration
Degeneration associated with Arthritic Disease is a known fact, as is the healing action of NBS. The customer database shows it acts on joints for easier movement and reduced stiffness long after the primary action of reducing inflammation has been achieved. In choosing the media for delivery of essential oils I wanted to break the mould of classic aromatherapy. This has turned out to be a clever choice as chemical analysis has now identified building blocks within other ingredients in the mix as compatible with the building blocks of bone, synovial membranes and cartilage. There has been interesting progress in the understanding of the collagen molecule. It is over 1400 amino acids long providing the sturdy structure for our bodies. Every third amino acid is glycine, a small amino acid that is abundant in NBS. Many of the remaining positions are filled by two unexpected amino acids one of which is also present in NBS: proline and a modified version of proline, hydroxyproline where we can also boast inclusion of the modification agent. Scientists didn't expect proline to be this common. It had been shown to form a kink in polypeptide chains that are difficult to accommodate in typical globular proteins so it has been of little interest in drug research. However it seems just the right structural shape for collagen.
Hydroxyproline is even more interesting. This amino acid is created by modifying normal proline after the collagen chain is built. That is unusual because it is critical for collagen stability and requires an agent that we cannot make ourselves and can only get from plants, vitamin C.
Theorisation of pharmacology relating to collagen.
By virtue of the direction that collagen is assembled, the primary structure is controlled by DNA at the N-terminal and progresses in sequence to the last amino acid at the C-terminal end. In nature additional amino acids are only added to the C-terminal end of proteins. The collagen chain is without biological activity. Function arises from conformation. The peptide unit is rigid and planar because of the triple bond character and there are no ionic receptors making it a sturdy protective barrier.
The structural flaw of collagen is its vulnerability to heat. On a molecular level, that is the energy released by oxygen reaction.
The existence of an everyday foodstuff Gelatin, shows the extent of collagen reaction to heat. Furthermore on cooling, collagen reacts with any available water to try and form a solid structure. In cooking, jelly. But in arthritis, globules that the body detects as undesirable and so the process goes on.
If arthritis is left untreated, the heat from inflammation causes lasting damage. As described previously, the cox-2 enzyme releasing oxygen indiscriminately has that burning effect. The resulting ionic charges from the structure damage attract other molecules that are detected as infection and so the process continues eroding more collagen. Taken to extreme, the membrane becomes so eroded that eventually the more stable permutation of collagen, your bones, are so close together that the triple helix can once again prevail. This in arthritis terms is "fusing of the bone".
NBS is able to form a bridge and restore the integrity of the helix for a short time. Repeated use maintains a stable environment for eventual healing
